Protein phosphorylation is the most common and important post-translational modification of proteins in the biological world. Since the 1950s, it has been regarded as a dynamic biological regulation process by biologists. About one-third of all proteins in cells are thought to be modified by phosphorylation. Protein phosphorylation is closely related to many biological processes, such as DNA damage repair, transcriptional regulation, signal transduction, and apoptosis regulation. The study of phosphorylated proteins and peptides can help us elucidate the mechanism of these processes and further understand the nature of life activities. With the development of proteomic technology in recent years, the research of protein phosphorylation has attracted more and more attention.

Phosphorylated polypeptide refers to the modification of the side hydroxyl group of Ser, Tyr and Thr residues in the peptide chain into acid phosphate polypeptide. Phosphorylated peptides are essential tools to study the phosphorylation process of proteins, so it is very important to study the phosphorylation reaction of proteins and peptides and determine the mature and simple synthesis route. So far, there are two main synthesis methods of phosphorylation and monomer. Post-phosphorylation method involves phosphorylation of Ser, Tyr or Thr side chain hydroxyl group after the polypeptide sequence is synthesized on the resin. The monomer method is the direct introduction of properly protected phosphorylated amino acids into the peptide sequence. This method is more convenient than the post-phosphorylation method, and has become the main method of peptide phosphorylation modification.
